Biological Activity

APL-4098 is an orally active, selective, ATP-competitive inhibitor targeting general control nonderepressible 2 (GCN2, also known as EIF2AK4). By inhibiting GCN2 kinase activity, APL-4098 impairs mitochondrial function and triggers the mitochondrial unfolded protein response. This pathway disruption leads to marked cytotoxicity in primary acute myeloid leukemia (AML) cells and leukemia stem cells. The GCN2-mediated stress response pathway is essential for maintaining cellular homeostasis and survival under nutrient deprivation and metabolic stress conditions. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
