Biological Activity
APG-2449 is an orally active small-molecule inhibitor of anaplastic lymphoma kinase (ALK), ROS proto-oncogene 1 receptor tyrosine kinase (ROS1), and focal adhesion kinase (FAK). By inhibiting ALK, ROS1, and FAK signaling pathways, APG-2449 suppresses cancer stem cell populations and cell proliferation, leading to potent and durable antitumor activity in non-small-cell lung cancer (NSCLC) and ovarian tumor models. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
