Biological Activity
Etrasimod (APD334) is a potent, selective, and orally available functional antagonist of the sphingosine-1-phosphate-1 (S1P1) receptor. By selectively engaging S1P1, etrasimod promotes receptor internalization and degradation, thereby disrupting the S1P-S1P1 signaling axis. Downstream, this inhibition impairs lymphocyte egress from secondary lymphoid organs into the circulation, attenuating immune cell infiltration and reducing neuroinflammation and autoimmune tissue damage in preclinical models. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
