Biological Activity
AMZ30 is a selective and covalent inhibitor of protein phosphatase methylesterase-1 (PME-1). By binding covalently to PME-1, AMZ30 inhibits its demethylase activity and prevents the demethylation of protein phosphatase 2A (PP2A). Consequently, AMZ30 reduces the demethylated form of PP2A in living cells, thereby restoring PP2A-mediated phosphatase functions and regulating downstream signaling pathways that control cellular proliferation and homeostasis. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
