Biological Activity

Amsacrine (m-AMSA) is a DNA-intercalating antineoplastic agent that acts as a potent inhibitor of topoisomerase II. By intercalating into DNA and stabilizing the topoisomerase II-cleavable complex, amsacrine blocks DNA strand re-ligation, leading to double-strand DNA breaks and inhibition of DNA replication. These molecular perturbations disrupt DNA synthesis, trigger cell cycle arrest, and induce apoptotic cell death in cancer cells. [1][2]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
