Biological Activity

AMG 925 is an orally bioavailable dual inhibitor targeting FLT3 (IC50 1 nM) and CDK4 (IC50 3 nM). By concurrently suppressing FLT3 and CDK4 signaling pathways, it blocks cell cycle progression at the G1 phase and downregulates downstream survival signaling. This dual mechanism leads to significant inhibition of cellular proliferation and induction of apoptosis in acute myeloid leukemia preclinical models. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
