Biological Activity

Sotorasib (AMG510) racemate is a racemic mixture of AMG-510, a potent and selective covalent inhibitor of KRAS G12C. By locking KRAS G12C in an inactive GDP-bound state, it inhibits downstream RAS/RAF/MEK/ERK signaling. This pathway blockade impairs cellular proliferation and induces apoptosis in KRAS G12C-mutated cancer cells, demonstrating strong antitumor activity in preclinical cancer models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
