Biological Activity
AMG-333 acts as a potent and selective antagonist targeting human and rat transient receptor potential melastatin 8 (TRPM8) ion channels. By blocking TRPM8 channel activation, AMG-333 inhibits menthol- and cold-induced calcium influx in primary sensory neurons, suppressing downstream thermal somatosensation and sensory signaling pathways. Blockade of this channel reduces cold allodynia and attenuates neuropathic pain responses in preclinical models. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
