Biological Activity

AMG 232 (Navtemadlin) is a selective inhibitor of the MDM2–p53 interaction that binds to MDM2 with a Kd ofand inhibits the interaction with an IC50 of. By disrupting this interaction, it reactivates the p53 pathway, leading to p53 stabilization, p21 expression, cell cycle arrest, and apoptosis in p53 wild-type tumor cells. It demonstrates robust in vivo antitumor activity across diverse xenograft models.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
