Biological Activity

Alvimopan dihydrate (LY246736 dihydrate) is a peripherally acting opioid antagonist targeting the μ-opioid receptor (Ki 0.77 nM), δ-opioid receptor (Ki 4.4 nM), and κ-opioid receptor (Ki 40 nM). It displays over 100-fold selectivity for opioid receptors relative to aminergic G-protein-coupled receptors. By selectively blocking peripheral μ-opioid receptors, alvimopan inhibits opioid-induced gastrointestinal hypomotility without compromising central analgesic signaling pathways. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
