Alrizomadlin (APG-115) is a highly potent small-molecule inhibitor of MDM2 that blocks the MDM2-p53 interaction. By preventing MDM2-mediated p53 degradation, it restores p53 pathway signaling and induces downstream p21 expression. This activation promotes cell cycle arrest and apoptosis in p53-wild-type cancer cells, resulting in potent antiproliferative activity and inhibition of tumor growth in xenograft models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
