Almorexant HCl is an orally active, dual orexin receptor antagonist that potently inhibits the OX1 receptor (IC50 13 nM) and OX2 receptor (IC50 8 nM). By blocking orexin peptide signaling, it suppresses central wakefulness pathways, leading to dose-dependent reductions in sleep latency and wake time after sleep onset. Preclinical studies in rats and dogs demonstrate sleep-promoting effects that resemble physiological sleep and are rapidly reversible upon sensorimotor stimulation. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
