Biological Activity

ALLO-2 is a potent antagonist targeting both wild-type mouse Smoothened (Smo) and the drug-resistant DH mutant human Smo. By binding Smo at a distinct site, it effectively blocks agonist-stimulated Gli reporter luciferase activity in TM-Gli-Luc cells. This suppression of the Hedgehog/GLI signaling pathway inhibits downstream Gli-mediated gene transcription, thereby blunting oncogenic signals and overcoming treatment resistance in Hedgehog-dependent cell models. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
