Biological Activity

Alkannin is a naphthoquinone derivative and potent, selective inhibitor of tumor-specific pyruvate kinase M2 (PKM2) that does not inhibit PKM1 or pyruvate kinase-L. By targeting PKM2, it blocks cancer cell aerobic glycolysis, resulting in reduced glucose consumption and decreased lactate production. This inhibition of glycolytic flux impairs cellular energy generation, suppressing proliferation and inducing cell death in drug-sensitive and resistant tumor cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
