Alcaftadine is a high-affinity histamine H1 receptor antagonist and an inhibitor of H2 and H4 receptors, while displaying no binding affinity for the H3 receptor. By antagonizing H1 and H4 receptor-mediated histamine signaling, it suppresses downstream inflammatory cascades, reduces vascular permeability, and prevents mast cell activation. These targeted pathway disruptions result in the inhibition of allergic conjunctivitis phenotypes and ocular itching.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
