Biological Activity

AKI603 is a small-molecule inhibitor of Aurora kinase A (AurA) developed to overcome imatinib resistance mediated by the BCR-ABL-TI mutation. In imatinib-resistant chronic myeloid leukemia cells, AurA inhibition by AKI603 suppresses cell proliferation and colony formation by inducing cell-cycle arrest and cellular senescence through elevated reactive oxygen species levels. Additionally, AKI603 attenuates tumor-initiating cell phenotypes in breast cancer models and downregulates stemness genes including β-catenin and c-Myc. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
