AGI-5198 is a selective inhibitor of mutant IDH1 R132H (IC50 = and IDH1 R132C (IC50 =. By targeting mutant IDH1 enzymes, AGI-5198 blocks the oncogenic production of R-2-hydroxyglutarate, leading to demethylation of histones and DNA. This inhibition promotes cellular differentiation, suppresses anchorage-independent colony growth, and delays tumor progression in preclinical glioma models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
