Biological Activity

AG 555 (Tyrphostin B46) is a tyrphostin-class synthetic inhibitor targeting epidermal growth factor receptor (EGFR) tyrosine kinase and topoisomerase I. By inhibiting EGFR kinase activity and directly interacting with topoisomerase I, AG 555 disrupts receptor autophosphorylation and prevents DNA relaxation. These combined actions block downstream oncogenic signaling cascades, leading to the suppression of EGF-dependent cellular proliferation and tumor growth. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
