AG-270 is a potent, non-competitive inhibitor of methionine adenosyltransferase 2A (MAT2A). By selectively blocking MAT2A activity, AG-270 depletes intracellular S-adenosylmethionine levels and disrupts essential downstream cellular methylation processes. This targeted inhibition effectively impairs cell survival and drives antiproliferative responses specifically in MTAP-deleted cancer cells, exploiting the synthetic lethal vulnerability associated with MTAP deficiency in tumors.
