AG-120 (racemic) is an orally active inhibitor targeting mutated isocitrate dehydrogenase 1 (IDH1). By suppressing mutant IDH1 enzymatic activity, the compound blocks the aberrant conversion of alpha-ketoglutarate to the oncometabolite 2-hydroxyglutarate (2-HG). Decreased intracellular 2-HG levels relieve epigenetic blockades, reversing histone and DNA hypermethylation to restore cellular differentiation and suppress tumor cell proliferation in mutant IDH1-driven malignancies. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
