Biological Activity

Afuresertib hydrochloride is an orally bioavailable, selective, and ATP-competitive pan-Akt kinase inhibitor targeting Akt1, Akt2, and Akt3 isoforms. By blocking Akt phosphorylation and activation, it disrupts downstream AKT/mTOR signaling pathways, leading to suppressed cell proliferation, cell cycle arrest, and the induction of apoptosis in tumor models. These molecular mechanisms contribute to its robust preclinical antitumor efficacy across various cancer models. [1][2]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
