Afimetoran (BMS- is an orally active, selective antagonist of Toll-like receptors 7 and(TLR7/. By inhibiting endosomal TLR7/signaling, it potently blocks NF-κB activation, attenuates pro-inflammatory cytokine secretion, and reverses TLR7-mediated steroid resistance in plasmacytoid dendritic cells. Furthermore, afimetoran suppresses disease activity in preclinical autoimmune models and reduces TLR7/pathway-associated gene expression in cutaneous lupus erythematosus. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
