Biological Activity

AF38469 is a selective, orally bioavailable inhibitor of sortilin with an IC50 value of 330 nM. By targeted inhibition of sortilin, AF38469 disrupts the binding and trafficking of key ligand partners such as neurotensin and proneurotrophins. This block in sortilin-mediated ligand internalization modulates downstream endosomal sorting pathways, altering lipid metabolism, lysosomal function, and neurotrophic factor signal transduction in preclinical models.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
