AF-353 is a potent, selective, and orally bioavailable antagonist of human P2X3 (p, rat P2X3 (p, and human P2X2/3 receptors (p. By selectively blocking P2X3- and P2X2/3-mediated purinergic signaling, AF-353 attenuates ATP-stimulated inward currents in sensory neurons. This blockade suppresses downstream nociceptive signal transmission, resulting in diminished pain behaviors and hyperalgesia in preclinical animal models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
