AC-4-130 is a selective inhibitor of the STAT5 SH2 domain that directly binds to STAT5. By disrupting the SH2 domain interaction, AC-4-130 blocks STAT5 tyrosine phosphorylation, dimerization, and subsequent nuclear translocation. This inhibition attenuates STAT5-dependent gene transcription, suppressing downstream oncogenic signaling pathways and reducing pathological STAT5-mediated cell proliferation in acute myeloid leukemia models.
