Biological Activity

AA92593 is a selective antagonist of melanopsin (OPN4) with an IC50 ofin a Ca2+ flux assay. By blocking OPN4-mediated signaling, it suppresses light-induced phototransduction in intrinsically photosensitive retinal ganglion cells (ipRGCs). Consequently, AA92593 inhibits downstream calcium influx and attenuates non-image-forming visual responses, providing a valuable tool for studying circadian photoentrainment and pupillary light reflexes. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
