A-366 is a potent, peptide-competitive inhibitor of G9a (IC50 3.3 nM) and GLP with >1000-fold selectivity over other methyltransferases. By inhibiting G9a and GLP catalytic activity, A-366 selectively decreases cellular global histone H3 lysine 9 dimethylation (H3K9me2) levels without significant acute cytotoxicity. This suppression of H3K9me2 alters chromatin structure, regulating gene transcription and impairing leukemia and cancer cell proliferation. [1]

- Preheat media/saline to 37°C.
