A-317491 is a potent and selective non-nucleotide antagonist of human P2X3 receptors (Ki = 22 nM) and human P2X2/3 receptors (Ki =. By inhibiting P2X3 and P2X2/3 receptor activation, it blocks purinergic agonist-evoked calcium influx and membrane currents in sensory neurons. This blockade attenuates peripheral nociceptive transmission, thereby reducing pain-related behaviors in preclinical models of chronic inflammatory and neuropathic pain. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
