Degrasyn (WP1130) is a selective deubiquitinase inhibitor targeting USP9x, USP5, USP14, and UCH37, while also suppressing JAK2/STAT and Bcr/Abl signaling without affecting 20S proteasome activity. By inhibiting tumor-activated deubiquitinases, WP1130 promotes downregulation of MCL-1 and upregulation of p53, thereby triggering aggresome formation, blocking autophagy, and inducing apoptosis in chronic myelogenous leukemia and other cancer cells. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
