Biological Activity
Tetrabenazine is a reversible high-affinity inhibitor of vesicular monoamine transporter 2 (VMAT2) that selectively blocks presynaptic monoamine uptake into granular vesicles. VMAT2 inhibition promotes early metabolic degradation of monoamines, particularly dopamine. Additionally, tetrabenazine exhibits antagonist activity at dopamine D2 receptors, albeit with lower affinity than for VMAT2. Depleting presynaptic dopamine stores suppresses downstream dopaminergic signaling and reduces hyperkinetic movement phenotypes. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
