SGI-1776 free base is an ATP-competitive, imidazo[1,2-b]pyridazine inhibitor of Pim family kinases, targeting Pim-1, Pim-2, and Pim-3, while also inhibiting FLT3 and Haspin. Inhibition of Pim kinases by SGI-1776 disrupts cell cycle progression, reduces anti-apoptotic Mcl-1 protein levels, and suppresses cell proliferation. Consequently, this leads to the induction of apoptosis and sensitization of cancer cells to cytotoxic chemotherapeutic agents. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
