Pyrimethamine is a dihydrofolate reductase (DHFR) inhibitor that targetedly suppresses folic acid biosynthesis pathways. By selectively inhibiting DHFR activity, Pyrimethamine blocks the reduction of dihydrofolate to tetrahydrofolate, depleting essential cofactors necessary for purine and pyrimidine nucleotide synthesis. This enzymatic inhibition downstream impairs DNA and RNA synthesis, thereby disrupting nucleic acid replication and suppressing cellular proliferation.

- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
