PluriSIn #1 is a selective inhibitor of stearoyl-CoA desaturase 1 (SCD1) that induces metabolic cell death in human pluripotent stem cells. By disrupting oleic acid synthesis and lipid homeostasis, SCD1 inhibition triggers endoplasmic reticulum stress, activates the unfolded protein response, and induces apoptosis specifically in undifferentiated stem cells. This selective cytotoxic effect depletes pluripotency-factor-expressing cells from heterogeneous populations, thereby preventing teratoma formation without compromising the survival of differentiated derivatives.
