Pizotifen Malate is a benzocycloheptane-derived antagonist primarily targeting serotonin receptors, including the 5-HT2 receptor subtypes, and histamine H1 receptors. By blocking serotonin and histamine receptor signaling pathways, it inhibits agonist-induced vascular smooth muscle contraction and suppresses neurogenic inflammatory processes. These actions modulate monoaminergic neurotransmission and downstream vascular tone regulation, leading to reduced cerebrovascular reactivity and inhibition of cranial vasodilation responses.
