Biological Activity

Rabusertib (LY2603618) is a potent and selective checkpoint kinase 1 (Chk1) inhibitor with antineoplastic activity. By suppressing Chk1 kinase activity, rabusertib impairs the DNA damage response checkpoint system and prevents the repair of damaged cellular DNA. This inhibition downstream leads to cell cycle arrest, persistent DNA double-strand breaks, induction of autophagy, and activation of Bak-dependent apoptosis, ultimately enhancing cancer cell sensitivity to DNA-damaging chemotherapeutic agents.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
