Haloperidol is a butyrophenone antipsychotic that functions as a dopamine receptor antagonist, targeting dopamine D2 receptors. By blocking post-synaptic dopamine D2 receptors, it inhibits G protein-coupled signaling pathways, resulting in altered cAMP accumulation and modulation of dopaminergic neurotransmission. These cellular actions attenuate hyperdopaminergic signaling and decrease spontaneous locomotor activity and psychotic-like phenotypes in neuronal and animal models.
