Glipizide is a second-generation sulfonylurea derivative that acts as a potent inhibitor of ATP-sensitive potassium (KATP) channels in pancreatic beta cells. By binding to the sulfonylurea receptor subunit and blocking these channels, glipizide induces cell membrane depolarization, opening voltage-gated L-type calcium channels. This promotes intracellular calcium influx and triggers the exocytosis of insulin storage granules to stimulate insulin secretion.

- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
Pre-aliquoted for Longevity: Solid-state aliquots offer significantly longer shelf life compared to stock solutions, ensuring maximum stability for every use.
