GSK872 is a potent and selective receptor-interacting protein kinase 3 (RIP3/RIPK3) inhibitor that binds directly to the RIP3 kinase domain. By inhibiting kinase phosphorylation within the RIPK1/RIPK3/MLKL signaling pathway, GSK872 effectively blocks necroptotic cell death. Consequently, it suppresses downstream necroptosis-associated phenotypes and RIP3-mediated inflammatory signaling responses, including interleukin-1β (IL-1β) maturation and cytokine secretion in immune cells.
