Biological Activity

CompK (compound K) is a potent and selective small molecule inhibitor of hematopoietic progenitor kinase 1 (HPK1 or MAP4K1). By inhibiting HPK1 within the T-cell receptor signaling pathway, CompK markedly enhances human T-cell immune responses under immunosuppressive conditions relevant to the tumor microenvironment, increasing T-cell receptor avidity to recognize tumor-associated antigens and boosting the cytolytic activity of CD8+ T cells. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
