Cladribine is a synthetic purine nucleoside analog and antimetabolite that targets proliferating and resting lymphocytes. Upon intracellular phosphorylation into its active triphosphate derivative, cladribine inhibits DNA synthesis and ribonucleotide reductase activity, inducing DNA strand breaks and activating apoptotic pathways. This mechanistically drives sustained cytotoxicity across hematologic cell lines, including multiple myeloma models such as U266, RPMI8226, and MM1.S. [1]

- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
