Calcifediol is a major circulating metabolite of vitamin D3 that targets the vitamin D receptor (VDR) signaling pathway. It suppresses parathyroid hormone (PTH) secretion and PTH mRNA expression in parathyroid cells, exhibiting an ED50 of 2 nM for PTH suppression. Through VDR activation, calcifediol regulates calcium homeostasis and bone mineralization pathways, reducing PTH levels to modulate mineral metabolism downstream. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
