Biological Activity

CUDC-101 is a potent multi-targeted inhibitor of HDAC, EGFR, and HER2. It effectively blocks class I and II HDACs and suppresses receptor tyrosine kinase autophosphorylation, leading to downstream inhibition of AKT and MAPK signaling pathways. Consequently, CUDC-101 exhibits robust antiproliferative activity across diverse cancer cell lines and induces marked tumor regression in xenograft models of non-small cell lung, pancreatic, and breast cancers. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
