CP-466722 is a potent and rapidly reversible ATM kinase inhibitor that exhibits selectivity over ATR and other PI3K-like protein kinase family members. By blocking cellular ATM kinase activity, CP-466722 inhibits ATM-dependent phosphorylation downstream in response to DNA damage, leading to cell cycle checkpoint defects. Furthermore, transient pharmacological inhibition of ATM with CP-466722 sensitizes cells to ionizing radiation, compromising clonogenic survival. [1]
