Biological Activity

Tosedostat (CHR2797) is an orally active aminopeptidase inhibitor targeting leucyl aminopeptidase (LAP), puromycin-sensitive aminopeptidase (PuSA), and aminopeptidase N (APN). Intracellular cleavage converts Tosedostat into its active metabolite, which inhibits target aminopeptidases and leads to cellular amino acid depletion. This disruption suppresses protein synthesis and downregulates the mTOR signaling pathway, resulting in cell cycle arrest and apoptosis in cancer cells. Furthermore, Tosedostat exhibits potent antiproliferative and antitumor effects both as a single agent and combined with chemotherapeutic drugs. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
