(E/Z)-BIX02188 is a potent and selective kinase inhibitor targeting MEK5 (IC50 of and ERK5 catalytic activity (IC50 of, while showing minimal activity against closely related kinases MEK1, MEK2, ERK2, and JNK2. By selectively suppressing the MEK5/ERK5 signaling axis, it blocks downstream transcription factor activation and inhibits gene expression associated with cell proliferation, vascular function, and neuronal responses. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
