AS-605240 is an orally active PI3Kγ inhibitor displaying 7.5-fold selectivity over PI3Kα and 30-fold selectivity over PI3Kβ and PI3Kδ. Inhibition of PI3Kγ suppresses PI3K/AKT signaling, thereby blocking neutrophil chemotaxis both in vitro and in vivo. Furthermore, AS-605240 treatment significantly reduces joint inflammation and minimizes the progression of joint destruction in mouse models of rheumatoid arthritis. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
