Biological Activity

5-Ethynyluridine (5-EU) is a cell-permeable nucleoside analogue and metabolic RNA labeler that is biosynthetically incorporated into newly synthesized RNA transcripts during transcription. Following incorporation into cellular RNA, 5-EU enables the detection and visualization of nascent RNA synthesis and turnover via copper-catalyzed azide-alkyne click chemistry cycloaddition. This selective metabolic labeling allows sensitive tracking of global transcription dynamics, gene expression changes, and RNA metabolic pathways without interfering with DNA synthesis. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
