Biological Activity

5-Chloro-2'-deoxyuridine (CldU) is a halogenated thymine analog and nucleoside antimetabolite used to investigate hypochlorous acid-induced DNA damage. Following intracellular phosphorylation, CldU is incorporated into DNA during replication, where 5-chlorouracil residues induce G:C to A:T transition mutations and prompt base excision repair pathways. Additionally, its monophosphate derivative inhibits thymidylate synthase, driving uracil incorporation into DNA and uracil-DNA glycosylase-mediated cytotoxicity.[1][2]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
