4EGI-1 is a small-molecule competitive inhibitor of the eIF4E/eIF4G interaction that directly binds to eIF4E. By disrupting the eIF4E/eIF4G complex, 4EGI-1 selectively inhibits cap-dependent translation initiation without altering cap-independent translation. This inhibition reduces expression of short-lived oncogenic proteins such as cyclin D1, resulting in cell cycle arrest and the induction of apoptosis in multiple cancer cell lines. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
