2-Thiouracil is a thiolated uracil derivative and antihyperthyroid agent that inhibits thyroid peroxidase to block thyroid hormone synthesis. By preventing the organification of iodide and coupling of iodotyrosines within thyroglobulin, it suppresses the production of triiodothyronine and thyroxine. This inhibition disrupts thyroid hormone-mediated signaling pathways, leading to altered metabolic regulation, reduced oxygen consumption, and decreased cellular metabolic rate. [1]
